چکیده

Brain edema after hypoxic-ischemic brain damage (HIBD) is a serious problem in neonates. Mannitol can mitigate edema; however, the molecular mechanism has not been fully elucidated. It has been reported that aquaporin-4 (AQP4), a member of the water channel protein family, plays a vital role in the development of brain edema. We therefore investigated whether mannitol might alleviate brain edema by regulating the expression of AQP4. In the present study, we used an established neonatal rat model of HIBD. A blood-brain barrier (BBB) permeability test revealed BBB leakage following HIBD, suggesting the vasogenic edema contributed to brain swelling during HIBD. Brain water content analysis indicated that mannitol could relief brain edema induced by HIBD after 6 h. In addition, qPCR showed that mannitol upregulated the expression of AQP4 in the first 48 h following HIBD induction. In conclusion, this study suggests that mannitol could alleviate brain edema after HIBD via upregulating the expression of AQP4. It may provide a novel insight into the treatment of HIBD.

Progesterone attenuates cerebral edema in neonatal rats with hypoxic-ischemic brain damage by inhibiting the expression of matrix metalloproteinase-9 and aquaporin-4

The aim of this study was to investigate the effects of progesterone (PROG) on blood-brain barrier (BBB) permeability, cerebral edema and the expression of matrix metalloproteinase-9 (MMP-9) and aquaporin-4 (AQP-4) in neonatal rats with hypoxic-ischemic brain damage (HIBD) and to explore the mechanism of its neuroprotective effect. Sixty 7-day-old Wistar rats were divided into sham surgery, hyp...

Role of melatonin receptors in the effect of estrogen on brain edema, intracranial pressure and expression of aquaporin 4 after traumatic brain injury

Objective(s): Traumatic brain injury (TBI) is one of the most common causes of death and disability in modern societies. The role of steroids and melatonin is recognized as a neuroprotective factor in traumatic injuries. This study examined the role of melatonin receptors in the neuroprotective effects of estrogen. Materials and Methods: Seventy female ovariectomized Wistar rats were divided in...

Goreisan Inhibits Upregulation of Aquaporin 4 and Formation of Cerebral Edema in the Rat Model of Juvenile Hypoxic-Ischemic Encephalopathy

Secondary cerebral edema regulation is of prognostic significance in hypoxic-ischemic encephalopathy (HIE), and aquaporin 4 (AQP4) plays an important role in the pathogenesis of cerebral edema. The traditional Japanese herbal medicine Goreisan relieves brain edema in adults; however, its effect and pharmacological mechanism in children are unknown. We investigated the effects of Goreisan on HIE...

Melatonin alleviates brain and peripheral tissue edema in a neonatal rat model of hypoxic-ischemic brain damage: the involvement of edema related proteins

BACKGROUND Previous studies have indicated edema may be involved in the pathophysiology following hypoxic-ischemic encephalopathy (HIE), and melatonin may exhibit neuro-protection against brain insults. However, little is known regarding the mechanisms that involve the protective effects of melatonin in the brain and peripheral tissues after HIE. The present study aimed to examine the effects o...

Effect of ischemic postconditioning on cerebral edema and the AQP4 expression following hypoxic-eschemic brain damage in neonatal rats.

BACKGROUND A rat model for neonatal hypoxic-ischemic brain damage (HIBD) was established to observe the effect of ischemic postconditioning (IPostC) on cerebral edema and the AQP4 expression following HIBD and to verify the neuroprotection of IPostC and the relationship between changes of AQP4 expression and cerebral edema. METHODS Water content was measured with dry-wet method, and AQP4 tran...
